I run a blank before each analytical sample (and therefore also between successive samples). I know from experience that my syringe rinsing routine prevents appreciable carryover between samples, but running a blank between each sample documents for each sample that there is in fact no carryover in that sample's analysis. That documentation is placed in the case file along with the analytical sample data. I use the same blank for each sample in a given batch of samples, and a fresh blank for each new batch. Sometimes I may use two blanks, if my batch of samples is so large that it would exhaust the contents of a single blank vial. My colleague instead places a separate blank between each pair of samples, such that half the samples in his sample rack are blanks, but he then reuses those blanks for several batches of samples. There is no significant advantage in doing it one way versus the other, but I see my method as providing a little extra insurance. The majority of my case samples are the same few drugs over and over again, so the possibility of carryover could easily be alleged if I didn't run blanks to prove carryover doesn't happen. After washing the autosampler syringe twice with solvent from each of my two rinse vials, the syringe is rinsed twice more with the solvent blank and then pumped twice back into the blank vial before drawing up the blank sample for actual injection (8 solvent rinses total, 6 ejected into the waste vial and the last two ejected back into the blank vial). Therefore, if there were even the most minute carryover occurring (so small that it didn't show up immediately), it would eventually build up in the single blank vial to the point where it would show up in one of the subsequent blank runs. The fact that I can run a large batch of many samples using that same single blank without any peaks ever showing up in any of the blank runs proves that all residual sample is eliminated by the wash routine BEFORE the sequence ever gets to the blank. Two of the columns are designed for the oven of the Agilent/HP 5890 GC, and were manufactured by Supelco. They are: 1 each - 6' glass column, ¼" OD, 0.2% Carbowax 1500 on 80/100 Carbopack C (a classic blood alcohol packing, but also suitable for other polar analytes) 1 each - 6' glass column, ¼" OD, 3% OV-1 on 100/120 Chromosorb WHP (a classic packing for general drug analysis, with many other applications) The other four columns were designed for the oven of the Beckman GC-65, a 1960's-1970's era floor-standing vertical GC. All were manufactured by Beckman. These are potential museum pieces, but are perfectly preserved (and who knows, there may still be a few of these GCs kicking around somewhere in the world). We found out that it's also an ASCLD issue, so we'll be running blanks after each sample from now on. If I may impose on the drug chemists one more time..... Does anyone have a procedure for extracting codiene from cough syrup? We are being asked to extract and quantitate codiene from these preparations, and we can't find a good procedure for this. The method was established in the 1960s and uses column chromatography (primitive, but should still work fine). Unfortunately, AOAC's copyright restrictions forbid my sending you a copy, but perhaps you have some other access to the Official Methods of Analysis of AOAC International. If not, the method cites JAOAC 42, 459(1959); 48, 607(1965) as references and perhaps you can get hold of them instead. Using 29 cranial measurements, the program does a linear discriminant analysis of an individual cranium. The result determines which of 66 groups of crania an individual is most like morphologically. The 66 groups comprise 2,870 crania from around the world. This database includes all the individuals that were in the database of W.W. Howells, but has additional samples from Europe, the Indian subcontinent, the Middle East and the extreme south of South America. The program comes with an illustrated manual that tells the user how to collect the measurements, run the program and interpret the results. The program runs under Windows. FORDISC utilizes modern forensic anthropological cases (you can see the breakdown in the 1998 Ousley & Jantz article "The forensic data bank: Documenting skeletal trends in the United States" in Forensic Osteology: Advances in the identification of human remains - Edited by Kathleen Reiches) from the US, and therefore should apply to US case work or you should make a reasonable argument to extend it to another population. FORDISC aslo includes post-cranial measurements to integrate into your analysis. CRANID picks up on measures of race/ethnicity only - but from around the globe. FORDISC only includes modern cases, so be forewarned when interpreting your data, and I cannot stress this enough, know the samples you are utilizing. The Physical Anthro section of AAFS is holding a session later this month in New Orleans on the use and abuse of FORDISC. You may want to check that out.. They are: 1 each - 6' glass column, ¼" OD, 0.2% Carbowax 1500 on 80/100 Carbopack C (a classic blood alcohol packing, but also suitable for other polar analytes) 1 each - 6' glass column, ¼" OD, 3% OV-1 on 100/120 Chromosorb WHP (a classic packing for general drug analysis, with many other applications) The other four columns were designed for the oven of the Beckman GC-65, a 1960's-1970's era floor-standing vertical GC. All were manufactured by Beckman. These are potential museum pieces, but are perfectly preserved (and who knows, there may still be a few of these GCs kicking around somewhere in the world). The problem comes about because of the way a spreadsheet program handles a comma-separated value file. What you do have to do is ensure that the decimal symbol for your system is set as a full-stop(.) not a comma. You do not have to change Regional and Language Options to English (Australia), though taking that more radical step has the same effect. You can do a quick check on your system's setting by opening the file CR5Form.CSV in your spreadsheet program. If the first cell in line 5 reads "GOL,?" then you have incompatible settings for the decimal symbol. The first cell in line 5 must read "GOL" and the second cell "?". How you reset the decimal symbol will vary from operating system to operating system. With Windows XP you go to Control Panel -> Regional & Language Options -> Regional Options -> Customize -> Decimal symbol. There select . instead of , and OK - > OK. You need only make these changes while you using your spreadsheet program (e.g. Excel) to make data files for CRANID. Once the CSV data file is created you can return to selecting a comma as your decimal symbol. CRANID itself is unconcerned about your Regional settings. Thank you again Dr Scatena.
